Okay, kind of a dumb question, but here goes:

If I’m cloning a partition doesn’t that mean I have to create a partition the same size as the existing one? IOW, would I have to create 1 partition the same size as the existing one (320GB), and then a 2nd one for the other 180GB?

Nope.
If you install Easeus on your current system, then shut down and attach the new drive in parallel with the old drive, boot to the old drive, the program will do everything including creating a partition. It can clone without using a bit by bit copy so the partitions can be different sizes.
