That would be The Rani, who was first seen teaming up with The Master in 1985’s “Mark of the Rani”.

(Before that, the Doctor had a Time Lady companion Romana during the last couple of years of Tom Baker’s era.

And of course, from the very first episode, we have Susan, as the Doctor’s grand daughter….

I could go on, but you probably get the point. The concept of female Time Lords is a no-brainer, since we were introduced to one in the very first episode before we even meet the Doctor. The only question was whether, in regeneration, there can be a change of what we might perceive as gender. And, given that regeneration involves a whole physical change of body, it’s not exactly a stretch, is it?)
